CRISPR/Cas9 technology is a powerful technique that has revolutionized the field of sequence-specific gene editing due to it's high precision, simplicity and flexibility. The CRISPR/Cas9 system consists of two major components: the Cas9 nuclease and the sequence-specific guide RNA (gRNA). They can form a very stable ribonucleoprotein (RNP) complex. Once a specific DNA region is matched by the gRNA carrying Cas9, the target DNA is cleaved with a double stranded break. Cas9 RNP complex can be introduced into cells directly and act immediately after it enters the cell without transcription and translation. By matching the gRNA sequence with different functional domains, the CRISPR/Cas9 system and its variants have been used for a broad range of gene editing applications (knock-out, knock-in, gene up/down, genome purification and visualization et. al.).

GenCrispr Cas9 Enzymes
DNA-free gene editing can be achieved by using purified Cas9 enzymes with gRNA and transfecting them directly into your cells or protoplasts of interest. GenCrispr Cas9 nuclease is the recombinant Streptococcus pyogenes Cas9 protein purified from E. coliand can be used for genome editing by inducing site-specific double stranded breaks in double stranded DNA. Cas9 protein forms a very stable ribonucleoprotein (RNP) complex with the guide RNA (gRNA) component of the CRISPR/Cas9 system. The RNP complex recognizes the target site by matching gRNA with the genomic DNA sequence and leads to DNA breaks within 3 bases from the NGG PAM (Protospacer Adjacent Motif). Presence of a nuclear localization signal (NLS) fusion with Cas9 enzyme ensures nuclear localization.
Enzyme activity validation guarantee
The GenCrispr Gene editing technology has been licensed-in from the Broad Institute. The activity of all GenCrispr Cas9 nucleases has been validated by in-vitro or in-vivo assays. The bioactivity data for all enzymes is included in the protocol document.
